SUMMARY:Back and forth between tumor initiation and tissue repair to dissect epidermal stem cell memory and plasticity
DESCRIPTION:Speaker: Giacomo Donati\, PhD – Associate Professor and Principal Investigator- Department of Life Sciences and Systems Biology\n& Molecular Biotechnology Center – University of Turin – Turin  | Italy. \nAbstract\n\nIt is well established that wound healing and epithelial cancer share a large set of features including immune cell infiltration\, extracellular matrix remodelling by fibroblasts\, induction of cell plasticity\, an epithelial-to-mesenchymal(-like) behaviour and enhanced cell motility. Moreover\, squamous cell carcinomas and epidermal cells involved in skin repair share a chromatin conformation state and a gene expression program\, if compared to homeostatic conditions. Recently\, we and others highlighted an important new link between wound healing and tumour initiation in epithelia. We showed that  epidermal cells undergoing an inflammatory event\, such as an injury\, acquire an epigenetic memory. This memory\, similarly to trained immunity in immune cells\, allows a more efficient response to a comparable future stimulus. Conversely\, when exposed to carcinogenic stimuli\, this wound memory exacerbated tumorigenesis. Overall\, we showed that this cell adaptation to injury relies on spatially organised memory-dedicated progenitors\, characterised by an actionable cell state that establishes an epigenetic field cancerization\, predisposing cells to tumour onset. In this talk\, I will discuss molecular and cellular features of this epithelial memory\, including its sub-organ spatial distribution. I will also present our new genetic approach to identify epigenetic regulators of epidermal stem cell plasticity.

DESCRIPTION:Barcelona Science Park and Wefern organize an event to present NEB. \nFor almost 50 years\, NEB has been a world leader in the discovery and production of reagents for the life science industry. New England Biolab’s enzymology expertise allows us to supply reagents for synthesis of high-quality RNA – form template generation to transcription\, to capping\, tailing and clean up after the synthesis. \nThe use of in vitro synthesized RNA has been instrumental in the development of RNA vaccines\, and CRISP/Cas9 genome editing tools\, generation of pluripotent stem cells\, screening of RNA inhibitors\, as well as development of RNA amplification-based diagnostics. DESCRIPTION:Speaker: Prof. Robert Tampé\, PhD – Professor of Biochemistry – Director of Institute of Biochemistry\, Biocenter – Goethe University Frankfurt\nFrankfurt | Germany. \nAbstract\n\nTo elicit an effective immune response against pathogens and cancerous cells\, major histocompatibility complex class I (MHC I) molecules undergo a complex journey from their biogenesis in the ER to their final decoding by cytotoxic T cells on target cell surfaces. Cellular machinery\, consisting of membrane transporters\, several chaperones\, and receptors\, facilitates MHC I assembly\, quality control\, and final recognition of peptide-MHC I. However\, the mechanistic integration of these processes remains poorly understood. I will discuss the multichaperone-client interaction network of the MHC I peptide loading complex assembled on the ABC transporter associated with antigen processing. Using integrative approaches\, we reveal the mechanistic underpinnings of antigen transport\, epitope proofreading\, quality control\, and the final release of MHC I complexes. Additionally\, I will present structural and mechanistic insights into the T cell receptor complex bound to a tumor-specific human class I pMHC. The T cell receptor complex is most fascinating due to its extraordinary ability to specifically recognize a vast array of antigens and its enigmatic signaling mechanisms that enable precise and highly regulated immune responses against cancer and pathogens.

DESCRIPTION:Servei de Fisioteràpia al Parc…ara també els dimarts! \nDavant la bona resposta i l’increment de demanda dels usuaris i les usuàries del PCB\, a partir del proper mes d’abril el Servei de Fisio s’amplia també als dimarts\,  amb el mateix horari. \nCost de l’activitat: Els imports són de 20€ per una sessió de 30 minuts i 36€ per una sessió de 60 minuts. El PCB cedeix gratuïtament una sala perquè un fisioterapeuta diplomat pugui fer les visites.
