SUMMARY:Disordered domains in oncogenic gene regulation

United States. \nIntrinsically disordered regions (IDRs) in DNA-associated proteins play important roles in oncogenic gene regulation but their overall genomic distribution and cooperative functions remain incompletely understood. In particular\, EWS fusion proteins\, which are formed by the combination of the EWS IDR and the DNA-binding domains of various transcription factors\, serve as paradigms for IDR function in cancer. Our laboratory has shown that the addition of the EWS IDR to the ETS transcription factor FLI1 in Ewing sarcoma has profound functional consequences\, leading to the formation of an oncogene that operates as a pioneer factor at new genomic locations that are not accessible to wildtype FLI1. Furthermore\, our data shows that EWS-FLI1 bound enhancers reconfigure the 3D nuclear architecture of Ewing sarcoma cells to establish an oncogenic gene regulation program that is critical for cell survival and proliferation. In order to identify other critical IDR containing proteins and characterize their roles in gene regulation programs\, we set out to develop methodologies to map the localization of DNA bound disordered proteins genome-wide. We recently reported a new antibody-independent chemical precipitation assay that can simultaneously map endogenous DNA-associated disordered proteins through a combination of biotinylated isoxazole precipitation and next-generation sequencing (DisP-seq\, disordered protein precipitation followed by DNA sequencing). DisP-seq profiles are composed of thousands of peaks that are associated with diverse chromatin states\, are enriched for disordered transcription factors (TFs) and are often arranged in large lineage-specific clusters with high local concentrations of disordered proteins and different combinations of histone modifications linked to regulatory potential. DisP-seq analysis of Ewing sarcoma cells revealed how disordered protein clusters containing EWS-FLI1 enable IDR-dependent mechanisms that control the binding and function of other disordered TFs\, including oncogene-dependent sequestration through long-range interactions and the reactivation of differentiation pathways upon loss of oncogenic stimuli.

SUMMARY:Ens hem de preocupar de la ciberseguretat?
DESCRIPTION:L’entitat de consultoria i serveis digitals avançats de la Universitat Politècnica de Catalunya (UPC) IThinkUPC i el Parc Científic de Barcelona organitzen el proper 26 de juny una jornada sobre ciberseguetat\, els riscos i amenaces que planteja i com afecta a les nostres organitzacions. El concepte ciberseguetat sembla ser tendència\, l’escoltem sovint als mitjans de comunicació\, a les converses i forma part d’algunes de les tasques que executem en el nostre dia a dia. Per què és tan important considerar la ciberseguretat? Aquesta xerrada analitzarà i intentarà fer entendre el context que aplica\, els riscos i amenaces que cal considerar\, i les raons. I acabarà per explicar quin és l’impacte de la ciberseguretat a la feina diària i com ens aplica i afecta per poder actuar en conseqüència i estar més preparats davant d’aquest gran repte.

SUMMARY:Servei de fisioteràpia per a la Comunitat del PCB
DESCRIPTION:Servei de Fisioteràpia al Parc…ara també els dimarts! \nDavant la bona resposta i l’increment de demanda dels usuaris i les usuàries del PCB\, a partir del proper mes d’abril el Servei de Fisio s’amplia també als dimarts\,  amb el mateix horari. \nCost de l’activitat: Els imports són de 20€ per una sessió de 30 minuts i 36€ per una sessió de 60 minuts. El PCB cedeix gratuïtament una sala perquè un fisioterapeuta diplomat pugui fer les visites. SUMMARY:Gene regulation of human cell systems
DESCRIPTION:Speaker: Roser Vento-Tormo\, PhD. Group Leader Cellular Genetics Programme – Wellcome Sanger Institute – Cambridge | UK. \nAbstract\n\nThe study of human tissues requires a systems biology approach. Their development starts in utero and during adulthood\, they change their organization and cell composition. Our team has integrated comprehensive maps of human developing and adult tissues generated by us and others using a combination of single-cell and spatial transcriptomics\, chromatin accessibility assays and fluorescent microscopy. We utilise these maps to guide the development and interpretability of in vitro models. To do so\, we develop and apply bioinformatic tools that allow us to quantitatively compare both systems and predict changes.
