DESCRIPTION:Speaker: Marc Liesa\, PhD – Group leader- Mitochondria\, redox and metabolic diseases – Institut de Biologia Molecular de Barcelona\, IBMB\, CSIC. \nAbstract:   \nMitochondria are commonly known as the powerhouse of the cell\, as they transform the energy contained in nutrients into ATP to fuel vital biochemical reactions. However\, mitochondria are not just powerhouses and they do not remain static and unchanged over time. Mitochondria have their own life-cycle\, with an incredible capacity to adapt to environmental stressors and generate signals that can model of the transcriptional landscape of a cell. Mitochondrial transporters and the interaction of mitochondria with other organelles have emerged as major mechanisms determining their bioenergetic and signaling activities to enable adaptation. In this talk\, I will discuss these mitochondrial mechanisms and how they control redox\, lipid metabolism and inflammation in health and disease.

Además\, también será un espacio de reflexión para profundizar en los últimos avances en el manejo clínico de la ataxia telangiectasia. \nEl evento\, organizado por el Instituto de Bioingeniería de Cataluña (IBEC) en colaboración con la AEFAT\, ofrecerá una variedad de actividades\, entre las cuales se destacan la presentación de los proyectos de investigación por parte de los propios investigadores\, y la participación de clínicos provenientes de diversos hospitales de Barcelona. \nEl presidente de la AEFAT clausurará la jornada y los asistentes podrán visitar las instalaciones de IBEC a fin de conocer mejor uno de los proyectos financiados. \nEl proyecto “Modelling Ataxia Telangiectasia pathogenesis and therapeutics using human pluripotent stem cells and genetic engineering”\, liderado por Núria Montserrat Pulido del Instituto de Bioingeniería de Cataluña (IBEC)\, tiene como objetivo establecer una plataforma celular para estudiar mediante ingeniería genética y celular fases tempranas de la ataxia telangiectasia. SUMMARY:Exploring the role of nucleolar stress in ageing and neurodegeneration
DESCRIPTION:Speaker: Dr. Vanesa Lafarga \nSenior Scientist\, Genomic Instability Group at CNIO (Spanish National Cancer Research Center)\, Madrid\, Spain. \nAbstract \nFor reasons that remain poorly understood\, alterations in ribosome biogenesis\, translation and nucleoli have a particular impact on neurons and hematopoietic cells. Notably\, mutations linked to neurodegenerative diseases are frequently related to nucleolar biology. A specific example of this is ALS (amyotrophic lateral sclerosis)\, a fatal neurodegenerative disease lacking a cure. The most frequent mutation in ALS patients is an intronic repeat expansion in C9ORF72\, which leads to the production of 2 types of arginine-rich dipeptide repeats (poly(PR) and poly(GR)) that cause nucleolar stress and cell death. We recently provided a mechanism that explains the toxicity of these peptides. By additional work\, we have now discovered that these peptides trigger an accumulation of free ribosomal proteins and mTOR hyperactivation\, a hallmark of ribosomopathies. In mice\, we see that the systemic expression of these peptides causes nucleolar stress and accelerated ageing\, which can be substantially alleviated by mTOR inhibition. In summary\, this work illustrates that the most frequent type of ALS might be a particular type of ribosomopathy. Furthermore\, our collective efforts\, as well as data coming from other groups\, suggests that the best approach to tackle neurodegenerative diseases might be to explore the value of strategies that can delay ageing as a whole. Our findings and ideas in this regard will be discussed.

SUMMARY:Regulating microtubule nucleation in neurons to control cell growth and polarity
DESCRIPTION:Speaker: Paul Conduit\, PhD.\nGroup Leader – Microtubule regulation in multi-cellular animals Lab – Institut Jacques Monod\, France. \nAbstract:   \nMy team aims to understand how microtubule nucleation is regulated across different cell types. The spatiotemporal control of microtubule nucleation allows cells to establish specialised microtubule networks that are necessary for a particular function\, such as separating chromosomes during mitosis or resisting compression during muscle contraction. Neurons are of great interest to us\, as they must generate and maintain extended microtubule networks that differ in polarity between axons (plus-end-out) and dendrites (mixed or minus-end-out). This polarity difference enables the correct trafficking of cargo into the different compartments and is therefore important for axon and dendrite identity and function. Microtubules are also important for neurite growth\, as they stabilise growth cones and support actin-dependent nascent neurite extensions. In this talk\, I will present our work using Drosophila dendritic arborisation (da) neurons as an in vivo model system. I will reveal how asymmetric microtubule nucleation within the soma is important to maintain minus-end-out microtubule polarity in proximal dendrites\, a mechanism likely conserved between different types of neurons. I will also show\, however\, that microtubule nucleation within dendrites is differentially regulated between different classes of da neurons to help establish class-specific dendritic arbor morphologies.

SUMMARY:Elucidating the molecular determinants of cell migration and virus assembly using cryo-electron tomography
DESCRIPTION:In this seminar I will give examples of how our work provided novel insights into the regulation of a key actin filament nucleator and the assembly of Vaccinia virus\, an important member of the poxvirus family. \nAbstract \n\nThe actin cytoskeleton defines emerging cellular properties\, including cell motility\, via a reciprocal crosstalk between actin filament geometry and actin binding protein (ABP) activity. Similarly\, virus assembly pathways are defined by geometrical principles that regulate virus particle architecture. The principles that govern the construction of pleomorphic viruses\, such as poxviruses or retroviruses\, show increased complexity due to their morphological variability\, where the local context of protein-protein interactions can have subtle but important effects on virus particle stability and function.\nThese examples of the actin cytoskeleton and virus assembly highlight that a full understanding of how higher-order protein interactions define emerging properties can only be obtained when they are studied within the appropriate contextual setting\, such as within the cell or assembled virus particles.\nIn our lab we develop novel approaches in cryo-electron tomography (cryo-ET)\, including specimen preparation\, data acquisition and image processing to study proteins in such complex environments and at different scales to integrate structural\, contextual\, and functional information.\nIn this seminar I will give examples of how our work provided novel insights into the regulation of a key actin filament nucleator and the assembly of Vaccinia virus\, an important member of the poxvirus family.

SUMMARY:TRNA epitranscriptomics as a new vulnerability in cancer
DESCRIPTION:Speaker: Dr. Pierre Close\, PhD. FNRS and WELBIO investigator & Head of the Laboratory of Cancer Signaling – GIGA-Institute \, CHU –Tour GIGA +2 | Liège\, Belgium. \nAbstract: \nTranslation reprogramming plays key roles in cancer development and drug resistance but the molecular mechanisms involved in this process remain poorly understood. Emerging evidences indicate that tRNA expression is modulated during tumorigenesis and correlates with changes in mRNA translation of genes with specific codon usage\, highlighting the importance of tRNA dynamics in cancer biology. Their recent work demonstrated that codon-specific mRNA translation reprogramming through regulation of tRNA modification promotes cancer development\, metastasis\, and resistance to targeted therapies. Specifically\, enzymes regulating wobble uridine tRNA modification are upregulated in several human cancers and they promote the expression of selected genes in a codon-specific manner. Their work highlights the importance of codon-specific mRNA translation and tRNA regulation as a source of future therapeutic targets in cancer.

DESCRIPTION:Servei de Fisioteràpia al Parc…ara també els dimarts! \nDavant la bona resposta i l’increment de demanda dels usuaris i les usuàries del PCB\, a partir del proper mes d’abril el Servei de Fisio s’amplia també als dimarts\,  amb el mateix horari. \nCost de l’activitat: Els imports són de 20€ per una sessió de 30 minuts i 36€ per una sessió de 60 minuts. El PCB cedeix gratuïtament una sala perquè un fisioterapeuta diplomat pugui fer les visites.
