SUMMARY:How in vitro multidrug resistance evolution studies with Candida auris not only provide insight in resistance mechanisms\, but also in possible treatment options
DESCRIPTION:Speaker: Prof. Patrick Van Dijck \nGroup Leader –  Molecular Biotechnology of Plants and Micro-organisms\, Department of Biology\, KU Leuven\, Leuven\, Belgium. \nAbstract \nThe increased prevalence of antifungal drug resistance and emergence of multidrug-resistant species such as Candida auris represent a global public health threat. We have setup high-throughput screenings using different clades of C. auris against a large set of antifungal drugs. Resistant isolates where then evolved for resistance against a second drug. Mapping drug susceptibility responses of such experimentally evolved C. auris strains across diverse antifungals\, resulted in combinations that show cross-resistance (XR) but also in combinatios that result in collateral sensitivity (CS).  This CS means that a strain that was resistant to drug A and is then evolved for resistance against drug B\, then becomes super sensitive towards drug A. Combinations showing CS were then validated using drug cycling and combination therapy\, providing a promising direction for future research. We also focused on the Amphothericin B resistance isolates in C. auris and obtained evidence for novel mechanisms of AmB resistance mechanisms and possible fitness compensation mutations.

Francisco Martínez-Jiménez will delve into the discoveries we’ve made and discuss how this knowledge can help us understand tumor evolution and develop better treatments for metastatic tumors.

SUMMARY:What can microfluidic technologies offer during self-assembly processes?
DESCRIPTION:Josep Puigmartí-Luis\, Departament de Ciència dels Materials i Química Física\, Institut de Química Teòrica i Computacional (invited by César Rodriguez-Emmenegger). \nSelf-assembly has long being used to control covalent and non-covalent interactions where molecular design has been the major driving force to achieve a desired outcome. Like in nature\, a full control over self-assembly processes could lead to rationalized structure-property correlations\, a long-time sought in chemistry\, physics and materials science. However\, the pathways followed and the mechanisms underlying the formation of supramolecular aggregates are still a major challenge for the scientific community. Accordingly\, the elucidation of nucleation and growth mechanisms will be highly required to push supramolecular chemistry to the next level\, where access to nature inspired functions will be accomplished. In this contribution\, I will present how reaction-diffusion (RD) conditions established within microfluidic devices can be used to uncover pathway complexity as well as to trigger pathway selection. Specifically\, I will show that microfluidic RD conditions provide an unprecedented kinetic control over self-assembly processes; for example\, enabling the isolation of well-defined kinetically trapped states as well as unprecedented metastable intermediates. This research provides a new tool to study and understand supramolecular chemistry\, and opens up new avenues for the engineering of advanced functional assemblies and systems.

DESCRIPTION:Speaker: Dr. Albert Cardona \nGroup Leader – Experimental and comparative connectomics\, Medical Research Council Laboratory of Molecular Biology (LMB)\, Cambridge Biomedical Campus\, Cambridge (UK). \nAbstract \nTo understand how the nervous system works\, we map its synaptic wiring diagram\, or connectome; optogenetically drive activity in select neurons to define a neuron-behaviour map; observe the activity of all neurons in a circuit; and study how neuromodulation restructures and repurposes circuits by changing neural circuit dynamics. All of the above is techonologically feasibly in small\, genetically tractable organisms such as the larval Drosophila melanogaster. Then\, on the basis of the connectome\, we formulate computational models of neural circuit function that can capture the observed activity dynamics that underlie behavior\, and which serves as a playground for formulating experimentally testable hypotheses of neural circuit function.

DESCRIPTION:Speaker: Maria Casanova-Acebes\, PhD. – Junior Group Leader – Cancer Immunity laboratory – Spanish National Cancer Research Center (CNIO) | Madrid\, Spain. \nAbstract: \nMyeloid cells dominate solid tumors yet due to their inherent plasticity; we have not succeeded at their effective targeting in cancer. This is worrisome because while novel immune checkpoint blockers arise in other immune lineages\, myeloid cells are in urgent need of effective therapies to dampen their immunosuppressive role in tumors. But well\, are these cells uniquely regulating immunosuppression in the tumor microenvironment (TME)? Can we functionalize all the efforts already made from single-cell technologies mapping this compartment? \nHer laboratory focuses on understanding HOW and WHEN myeloid cells become relevant in the TME of solid tumors. By asking how they crosstalk with key cellular and non-cellular players in transformed tissues\, they aim to identify effective and durative ways of impeding their pathogenic program in tumors. During her talk\, She will go over some published (to set the stage) and unpublished data in which: 1) I will pitch on the role of alveolar macrophages modulating the stroma of lung tumors; 2) provoke the audience with exciting data on circadian rhythms in the lung TME (and how myeloid cells modulate those!->this is our WHEN angle) and finally\, 3) introduce exciting data on how clonal hematopoiesis impacts the outcome of ovarian tumors and macrophage inflammatory and lipid metabolic activity.

SUMMARY:Lead Generation without an X-Ray Crystal Structure: An NMR Method to Probe Protein-Ligand Complexes
DESCRIPTION:Speaker: Prof. Julien Orts. University of Vienna\, Faculty of Life Sciences\, Department of Pharmaceutical Sciences\, Austria. \nAbstract \nX-ray crystallography molecular replacement (MR) is a highly versatile tool for the detailed characterization of lead compound and binding modes in the pharmaceutical industry. The two major limitations of its application to drug research are (i) the availability of a similar protein structure\, and (ii) obtaining well-diffracting crystals of the ligand-protein complexes of interest. While nowadays the first point is often not a limitation anymore\, obtaining well-diffracting crystals may be difficult. In such situations structure determination of protein-ligand complexes by liquid-state NMR is a good option. Unfortunately\, the established standard structure determination protocol is in general time-consuming\, and a shortcut using available structural data as in the case of MR in X-ray crystallography is not available.\nHere\, we present NMR2 (NMR Molecular Replacement)\, a MR-like approach in NMR to determine the structures of the binding pockets of ligands at atomic resolution. The calculation of structures of protein-ligand complexes relies on the collection of unassigned semi-quantitative inter-molecular NOE distance restraints and on previously solved structures. The NMR2 method uses a high throughput structure calculation protocol\, rather than a docking-scoring simulation. It is fast since it requires only a few days of measuring time and bypasses the time-consuming sequential assignment steps for the protein.\nWe will present multiple NMR2 applications covering several ligand topologies ranging from peptidomimetic to small molecules that bind strongly or weakly to protein receptors. We also report how NMR2 can make use of partially labelled protein using methyl-specific isotope labelling. Finally\, we will present our latest methodology development to further advance the technique. Our findings demonstrate that NMR2 may open an avenue for the fast and robust determination of the binding pocket structure of ligand-protein complexes at atomic resolution.

SUMMARY:Peptide Therapeutics for Gastrointestinal Disorders

IRB Barcelona Alumni Award 2022. \nAbstract: \nPeptides have emerged as a therapeutically and commercially important drug class owing to their exceptional specificity\, potency and low toxicity. They also serve as invaluable research tools\, enabling the investigation of the physiological functions of receptors and underlying\nmechanisms of various diseases. The profound impact of peptide research is evident with the approval of over 80 peptide therapeutics approved and more than 150 peptides undergoing clinical trials\, supporting innovative treatments for numerous conditions\, including diabetes\, cancer\, multiple sclerosis\, gastrointestinal disorders\, obesity and pain. \nA main limitation of peptides is their intrinsic susceptibility to proteolytic degradation and lack of oral bioavailability. Nevertheless\, several peptide families exist that are produced and function effectively in the gastrointestinal tract\, arguably one of the most hostile environments for peptides. Our interest lies in understanding the structural makeup of these peptides to comprehend how they can function in such an environment and to utilise this knowledge for the development of gut-specific peptide therapeutics. \nOne intriguing group we study is the intriguing trefoil factor family (TFF) which displays a rigid disulfide bond-stabilised multiloop structure reminiscent of a trefoil. TFF peptides play a crucial role in gastrointestinal protection and repair\, offering promising therapeutic potential for the treatment and prevention of gastrointestinal disorders. \nIn addition\, we have systematically evaluated other naturally occurring peptide scaffolds and medicinal chemistry approaches to identify new strategies for the development of oral and gut-specific peptide therapeutics.\nDrawing upon these studies\, we have successfully developed several oral therapeutic lead candidates addressing unmet medical needs\, such as chronic abdominal pain\, gastrointestinal biofilms\, and irritable bowel syndrome. These fundamentally new approaches represent a major step forward in the field of peptide drug development and hold great promise for improving the lives of patients suffering from chronic gastrointestinal disorders.

El PCB cedeix gratuïtament una sala perquè un fisioterapeuta diplomat pugui fer les visites.
