SUMMARY:The causation of childhood leukemia
DESCRIPTION:Speaker: Isidro Sanchez-Garcia\, MD & PhD – Experimental Therapeutics and Translational Oncology Program\, Instituto de Biología Molecular y Celular del Cáncer – CSIC/ Universidad de Salamanca. Spain. \nAbstract: \nIn recent years\, the application of comprehensive genomic approaches has revealed a plethora of germline and somatic genetic variants that increase cancer risk significantly for both hematopoietic and solid malignancies. In contrast to the long-standing view that germline predisposition affects young people preferentially and biases to the development of solid tumors\, recent data from the study of hematopoietic malignancies indicate that germline risk exists throughout the entire age range of life and the pathways involved overlap with those that drive solid tumors. Mutations driving cancer in adults can be inherited or even occur at the time of fertilization as de novo mutations\, and thus\, can be established in utero\, and not later in life. Moreover\, the acquisition of somatic mutations in hematopoietic tissues\, now referred to as clonal hematopoiesis\, itself provides independent risk for the subsequent development of both hematopoietic as well as solid tumors\, with inflammatory signaling likely to be of critical importance. Thus\, we are developing a much more cohesive view of cancer risk with paradigms that are unifying across age and tumor spectra. The amassing information is providing unique insights into the etiology of tumor formation and informing approaches to clinical care. Accordingly\, the field of genetic predisposition to cancer is advancing rapidly and affecting therapeutic decision-making and prophylactic measures for affected patients\, children\, and other close relatives. Here\, I will provide insights into how genetic and environmental factors interact to influence predisposition to acute leukemia in children versus adults and discuss how this knowledge can impact prevention strategies.

SUMMARY:Staying fit for the next generation: suppression of mitochondrial complex I in oocytes
DESCRIPTION:Speaker: Dr. Aida Rodríguez-Nuevo \nSenior Postdoctoral Researcher\, Oocyte Biology & Cellular Dormancy Group\, Centre for Genomic Regulation (CRG)\, The Barcelona Institute of Science and Technology (BIST). \nAbstract \nThe conservation of the germline becomes essential when zooming out from cell biology into species fitness. Oocytes\, the female germ cells that become eggs\, form before birth and remain viable for several decades before fertilization. Yet\, little is known about the mechanisms employed by oocytes to deliver a pristine cytoplasm to the next generation\, after decades of being formed. We discovered that oocytes evade the production of reactive oxygen species by remodelling the mitochondrial electron transport chain through elimination of complex I. Combining live-cell imaging and proteomics in human and Xenopus oocytes\, we find that early oocytes exhibit greatly reduced levels of complex I. This is accompanied by a highly active mitochondrial unfolded protein response\, which is indicative of an imbalanced electron transport chain. Biochemical and functional assays confirm that complex I is neither assembled nor active in early oocytes. Thus\, we report a physiological cell type without complex I in animals. Complex I suppression represents an evolutionarily conserved strategy that allows longevity while maintaining biological activity in long-lived oocytes. \n

DESCRIPTION:Desde Agilent nos gustaría invitarle al evento organizado por las divisiones de Cell Analysis y Genómica de Agilent llamado “ Uso de las soluciones de Agilent Technologies y exploración de las tecnologías y aplicaciones disponibles para el día a día en el laboratorio” que tendrá lugar el lunes 23 de Octubre. El seminario será impartido por Miriam Contreras\, responsable de Cell Analysis en Agilent España\, Yolanda Sanchez\, responsable de Genómica en Agilent España\, y Carles Ramos\, Inside sales de Cell Analysis en Agilent España. \nEl objetivo de este Seminario/demo es mostrar todas las tecnologías y aplicaciones de esas tecnologías que creemos que puede ser de su interés:\n• Dar soluciones para la investigación o desarrollo que lleven a cabo\n• Mostrar nuevas aplicaciones/instrumentos que no conocían y que les\npodrían ser útiles\n• Establecer un lugar para comunicar y hablar de las necesidades de cada\nusuario y ver si en algún caso es interesante alguna colaboración en el uso\nde equipos o compartir una compra\, etc. \nEl seminario tendrá una duración de dos horas aproximadamente y después estaremos a su disposición para atender sus consultas y dudas.

Universidade de Santiago de Compostela\, Spain. \nAbstract \nThe development of artificial metalloenzymes (ArMs) that catalyze novel transformations and expand the repertoire of biocompatible reactions available in living cells is a major research challenge at the intersection of catalysis\, protein engineering\, and chemical biology. Catalytic metallopeptides\, halfway between conventional transition metal catalysts and protein-based ArMs\, may hold the key to efficient catalysis in biological settings. In this context\, we will describe the latest advances in our group in the development of catalytic metallopeptides using rational methods and combinatorial chemistry.

SUMMARY:Cell To Cell Communication In Glioblastoma Progression
DESCRIPTION:Speaker: Sergio Casas Tintó\, PhD – Instituto de Investigación de Enfermedades Raras (IIER-AGH) – Unidad de Enfermedades Humanas en Drosophila – Instituto de Salud Carlos III – Madrid. \nAbstract: \nGlioblastoma (GB) is the most aggressive\, lethal and frequent primary brain tumor. It originates from glial cells and is characterized by rapid expansion through infiltration. We have studied how GB cells interact with the microenvironment and healthy surrounding tissues\, mostly neurons. GB cells project tumor microtubes (TMs) that contact with neighboring neurons\, and exchange signaling molecules related to Wingless/WNT\, JNK\, Insulin or Neuroligin-3 pathways. This cell to cell communication promotes GB expansion and synapse loss in the neurons. Moreover\, healthy neurons form glutamatergic functional synapses with GB cells which facilitate GB expansion and premature death. Targeting signaling and synaptic components of GB progression may become a suitable strategy against glioblastoma. In a Drosophila GB model\, we have determined the post-synaptic nature of GB cells with respect to neurons\, and the contribution of post-synaptic genes expressed in GB cells to tumor progression. In addition\, we document the presence of intratumoral synapses between GB cells\, and the functional contribution of pre-synaptic genes to GB calcium dependent activity and expansion. Finally\, we explore the relevance of synaptic genes in GB cells to the lifespan reduction caused by GB advance. Our results indicate that both presynaptic and postsynaptic proteins play a role in GB progression and lethality.

SUMMARY:Relevance of circadian physiology for disease and aging
DESCRIPTION:Speaker: Amita Sehgal\, PhD. – John Herr Musser Professor of Neuroscience – Vice Chair\, Department of Neuroscience – Director\, Chronobiology Program\, Perelman School of Medicine\, University of Pennsylvania | Philadelphia\, United States. \nAbstract: \nCircadian (~24 hour) rhythms of behavior and physiology are generated by endogenous clocks located within tissues of almost all organisms. We now have some understanding of how these clocks are generated and how they synchronize to environmental signals. However\, mechanisms by which clocks drive rhythms of downstream processes\, and how disruption of clocks contributes to pathological conditions are not well understood. We find that the circadian clock drives rhythms in components of the cell cycle\, which provides a mechanism for effects of circadian disruption on cancer progression. A chemotherapeutic drug targeted to such a rhythmic component has time-of-day specific effects in cells and in a mouse tumor model. In fact\, we find that\, of 126 anti-cancer drugs we screened\, ~half work better at a specific time of day. As many of these temporal effects require clocks within tumors\, we have also addressed the question of why some tumors have clocks while others don’t. In other work\, to address the relationship between aging and circadian disruption\, we have investigated how circadian regulation of downstream genes changes with age.
