DESCRIPTION:El Parc Científic de Barcelona y Paralab Bio organizan el seminario Microscopía Intravital en Tiempo Real (IVIM): Imágenes a nivel celular de órganos internos en un animal vivo para explicar esta interesante técnica. \nLa microscopía intravital es una fusión entre las disciplinas de experimentación animal/pre-clínica y la microscopía óptica avanzada. Por ello\, es la única técnica que permite la visualización a nivel microscópico in vivo en organismos modelos\, como el ratón. \nEl desarrollo de este tipo de equipo habitualmente ha sido responsabilidad de responsables de servicio y usuarios. IVIM technologies\, representado por Paralab Bio en Iberia\, otorga a los investigadores un equipo desarrollado específicamente para microscopía intravital optimizado para visualizar con múltiples accesorios validados experimentalmente y estrategias experimentales para obtener resultados en una gran multitud de órganos. \nNo os perdáis la oportunidad de conocer la tecnología detrás de este equipo y de poder repasar los papers peer reviewed que han realizado otros usuarios utilizando esta tecnología en su investigación.

SUMMARY:Deciphering the cis-regulatory code that guides development using interpretable deep learning
DESCRIPTION:Speaker:   Dr Julia Zeitlinger\, Investigator at the Stowers Institute for Medical Research and Associate Professor at the University of Kansas Medical Center. \nAbstract:   \nThe cis-regulatory code that instructs gene regulation during development\, also known as the genome’s second code\, is a fundamentally unresolved problem. Recent progress has provided proof-of-principle evidence that this complex cis-regulatory code can be learned with neural networks. The new approach is fundamentally different from traditional methods in that the sequence rules are learned inside a black box directly from genomic sequences through their ability to better predict a specific genomics data set. This dramatically improves the predictive performance\, but requires rigorous approaches for extracting\, understanding and validating the learned sequence rules to make sure that they represent biology. I will describe how we use this approach using mouse or Drosophila development as model systems and uncover unexpected sequence rules that we can validate with experiments.

SUMMARY:Insights into lung cancer evolution and metastases in TRACERx and PEACE

Group Leader – Cancer Metastasis Laboratory – CRUK Lung Cancer Centre of Excellence\, UCL Cancer Institute | London\, United Kingdom. \nAbstract: \nTRACERx is a prospective longitudinal cancer evolution programme which investigates the relationship between intratumour heterogeneity and clinical outcomes in non-small cell lung cancer (NSCLC). Early findings from the study demonstrated the prognostic relevance of increased chromosomal\, as opposed to mutational\, heterogeneity\, as well as the clinical utility of circulating tumour DNA (ctDNA) in tracking cancer evolution. Heterogeneity in gene expression was found to identify a cohort of patients with early stage I NSCLC with high risk disease\, suggesting a role for adjuvant therapy in patients who would otherwise not receive it. Tumour immune evasion was prevalent and prognostically relevant in treatment-naïve NSCLCs\, and neoantigen heterogeneity and clonality were adopted to establish early phase studies in adoptive T cell therapies.\nIn the current cohort of 421 patients with extended follow up\, matched primary and metastatic analysis has led to further findings in relation to impact of cancer evolution on clinical practice and patient outcomes. Large dominant subclones in primary tumours were associated with positive subclonal selection\, and mutations in specific genes were found to be under significant subclonal\, and not clonal\, selection. Subclonal genome doubling was identified in 20% of tumours and multiple genome doubling events were identified in 10% of tumours. Patients with tumours containing large recent subclonal expansions had significantly shorter disease-free survival (DFS)\, and as previously demonstrated\, chromosomal instability was associated with poor outcomes\, particularly\, extrathoracic disease relapse.\nMetastatic relapse was found to occur late in tumour evolution (late divergence; after the last clonal sweep in the primary tumour) in 75% of tumours\, and early divergence (25% of tumours) was found to occur at predicted tumour volumes below the threshold for further investigation using radiological imaging\, suggesting potential implications for clinical intervention. Metastasis-seeding subclones in primary tumours were subject to subclonal expansion\, likely reflective of the acquisition of subclonal driver mutations and somatic copy number alterations. ctDNA analyses in this larger cohort demonstrated the value of ctDNA to predict the likelihood of future relapse\, but also its use in tracking tumour growth dynamics and emergence of relapse. By integrating histological growth patterns with tumour genomics in lung adenocarcinoma\, predominantly high-grade patterns were associated with increased chromosomal complexity\, subclonal somatic copy number alterations and loss of heterozygosity.  Specific copy number alterations were found to be associated with high- and low-grade patterns\, suggesting distinct evolutionary trajectories according to growth patterns. The presence of specific growth patterns combined with other histological features and ctDNA identified patients with an increased risk of extrathoracic recurrence\, suggesting a potential role for both morphological and genomic subtyping to aid risk stratification

SUMMARY:Disrupting Metabolic Support Networks in Pancreatic Cancer
DESCRIPTION:Speaker: Christopher J. Halbrook\, PhD – Assistant Professor\, Molecular Biology and Biochemistry. School of Biological Sciences – University of California\, Irvine. Irvine\, USA. \nAbstract: \nPancreatic ductal adenocarcinoma (PDA) remains one of the deadliest major cancers. A key barrier to PDA treatment is the physiology of the tumors\, characterized by a densely fibrotic extracellular matrix deposited by fibroblasts\, rich with immune cell infiltration. The diversity of epithelial\, stromal\, and immune cell populations within pancreatic tumors provides numerous avenues supporting cancer growth and survival. Our work has shown that disruption of metabolic crosstalk between different tumor compartments functions to impair tumor growth\, reduce immune suppression\, and potentiate response to chemotherapy.\nRecently\, we have identified an axis of crosstalk between clonal populations of cancer cells driven by a differential integrated stress response. Targeting the resulting nutrient exchange by degrading exogenous asparagine sensitizes pancreatic tumors to mitochondrial inhibition. Our current efforts aim to characterize how this treatment remodels PDA tumors in both primary and metastatic disease models and determine the pre-clinical utility of this approach. While these seminal studies only begin to describe the metabolic interactions shaping the microenvironment\, they demonstrate the potential of uncoupling metabolic symbiosis to target pancreatic cancer.

DESCRIPTION:Servei de Fisioteràpia al Parc…ara també els dimarts! \nDavant la bona resposta i l’increment de demanda dels usuaris i les usuàries del PCB\, a partir del proper mes d’abril el Servei de Fisio s’amplia també als dimarts\,  amb el mateix horari. \nCost de l’activitat: Els imports són de 20€ per una sessió de 30 minuts i 36€ per una sessió de 60 minuts. El PCB cedeix gratuïtament una sala perquè un fisioterapeuta diplomat pugui fer les visites.
