SUMMARY:Regulation of Hematopoietic Stem Cell Dormancy

Principal Investigator\, Center for Integrative Biological Signaling Studies (CIBSS)\, German Cluster of Scientific Excellence\, University of Freiburg. \nAbstract: \nBone marrow hematopoietic stem cells (HSCs) are vital for the maintenance of hematopoiesis. In inbred mice housed in gnotobiotic facilities\, the top of the hematopoietic hierarchy is occupied by dormant HSCs\, which reversibly exit quiescence during stress. Still\, whether HSC dormancy exists in human remains unclear. Our recent work shows the existence and relevance of dormancy in human HSCs and identifies the hyaluronic acid-GPRC5C signalling axis as an essential component controlling the HSC state (Zhang et al.\, Nature Cell Biology 2022). Further\, HSCs rely on complex regulatory networks to preserve their function. Due to the scarcity of HSCs\, technical challenges have limited their insights into the interplay between HSC metabolism and their transcriptional and epigenetic regulation. They recently established and integrated low-input multi-layer OMICs data revealing distinct metabolic and epigenetic hubs that are enriched in HSCs and their downstream multipotent progenitors (Schönberger et al.\, Cell Stem Cell 2022). Mechanistically\, they uncover a non-classical retinoic acid signaling axis that regulates HSC identity. Their findings emphasize how a single metabolite controls stem cell fate by instructing epigenetic and transcriptional attributes.

Team Leader\, Imaging the Cardiovascular Inflammation and the Immune Response\, Spanish National Cardiovascular Research Center (CNIC)\, Madrid\, Spain. \nInnate immune cells are there to afford protection to the host in an environment that is full of opportunistic pathogens. Or perhaps a better definition is that innate immunity is an extremely adaptable system that facilitates adaptation to an ever changing environment\, be it due to infections\, age\, or other challenges. In his lab\, they study the rules of these adaptations by focusing on two major innate immune cells\, macrophages\, and neutrophils. He will present here the case of neutrophils\, the new biology that they and others are discovering as they visit different tissues\, and the opportunities that this knowledge opens in biomedical research.

SUMMARY:Accelerating drug discovery using advanced protein and antibody development platforms
DESCRIPTION:Sino Biological and Barcelona Science Park organize an event to present different strategies to accelerate drug discovery using advanced protein and antibody development platforms. \nCancer is a disease characterized by misregulation of cell proliferation and differentiation. Moreover\, cancer cells often escape the immune system’s attacks by expressing inhibitory ligands and molecules. Immunotherapies such as immune checkpoint blockade\, chimeric antigen receptor T-cell (CAR-T) therapies\, antibodies to cancer antigens and cytokine therapy overcome the immune escape and offer a promising treatment for several cancers\, as well as autoimmune diseases. Therefore\, manufacturing high-quality therapeutic proteins and antibodies has become very important to meet medical needs. However\, low protein yield\, protein aggregation and degradation are common issues during therapeutic-protein production. \nSino Biological has provided many strategies to solve these difficulties such as codon optimization\, single amino acid mutation and improvement of protein purification. Using several antibody development platforms and different protein production methods\, Sino Biological has generated many therapeutic antibodies such as neutralization antibodies against SARS-CoV-2 or RSV; several immune checkpoint blockade antibodies including anti-PD-1\, anti-CTLA-4 and anti-TIM-3 antibodies. Utilizing comprehensive cell-based assays\, they have demonstrated therapeutic functions of these antibodies in vitro. Additionally\, they have developed many long half-life therapeutic cytokines. Combining these cytokines with immune checkpoint blockade has shown synergistic effects to inhibit cancer progression\, which can be applied on the preclinic trials and are benefits for cancer treatment in vivo.

DESCRIPTION:Desde FI Group\, te invitamos a participar de la mano de CDTI y ASEBIO\, en una sesión informativa que tendrá lugar el 23 de Marzo a las 11h de manera presencial en el Parc Científic de Barcelona. \nEn la jornada se tratarán los principales retos tecnológicos del sector biosalud y farmacéutico\, así como se hablarán de las distintas ayudas para la transferencia tecnológica de proyectos dentro del ámbito sanitario. Las principales convocatorias de ayudas que tratará CDTI estarán orientadas hacia los siguientes programas: \n\nInstrumentos CDTI en los que se promueve transferencia tecnológica a través de proyectos I+D: principalmente programa CIEN que promueve la transmisión biotecnológica entre empresas y organismos de investigación y proyectos de cooperación nacional e internacional; instrumentos enfocados para pymes y grandes empresas.\nNeotec: orientado a startup para promover la transferencia de las investigaciones\, y apoyar la creación y consolidación de empresas  de base tecnológica y de reciente creación.\nInnvierte CDTI: para apoyar y promover empresas innovadoras a través de la entrada de capital.\nCompra pública pre-comercial del CDTI\, enfocada en el área de salud\, sobre la transferencia de tecnología para proporcionar prototipos que respondan a las necesidades de administraciones públicas no cubiertas.\nProyectos Transferencia Cervera*: ayudas a proyectos de I+D para favorecer la cooperación entre las empresas y organismos públicos\, instrumento pendiente de publicación\, con reformulación de condiciones de presentación.

SUMMARY:Decoding and Intercepting ER Stress Signals for Cancer Immunotherapy

Associate Professor of Immunology and Microbial Pathogenesis. Co-Leader\, Cancer Biology Research Program. \nMeyer Cancer Center. Weill Cornell Medicine – Cornell University. New York\, USA. \nThe tumor microenvironment provokes endoplasmic reticulum (ER) stress in malignant cells and infiltrating immune populations. Sensing and responding to ER stress is coordinated by the unfolded protein response (UPR)\, an integrated signaling pathway governed by three ER stress sensors: ATF6\, IRE1α\, and PERK. Dysregulated UPR activation in cancer has been shown to dictate malignant progression\, tumor growth\, metastasis\, and protective anti-tumor immunity. Hence\, therapies targeting the UPR can be harnessed to elicit direct tumor killing and concomitant anti-cancer immunity. This seminar highlights multiple research efforts from our laboratory that revealed the key immunomodulatory function of ER stress signaling in cancer.

SUMMARY:Quantitative microbiome profiling in health and disease
DESCRIPTION:Speaker:  Prof. Jeroen Raes\, Phd – Group Leader Bioinformatics and (Eco)-Systems Biology Lab – Department of Microbiology and Immunology\, Rega Institute for Medical Research\, KU Leuven Belgium – Vice- Director VIB Center for Microbiology Belgium. \nAbstract: Alterations in the gut microbiota have been linked to various pathologies\, ranging from inflammatory bowel diseases and diabetes to cancer.\nAlthough large numbers of clinical studies aiming at microbiome-based disease markers are currently being performed\, our basic knowledge about the normal variability of the human intestinal microbiota and its determining factors remains limited. Here\, I will discuss our findings studying a large-scale study (Flemish Gut Flora Project; n=3400) of the gut microbiome variation in a geographically confined region (Flanders\, Belgium)\, in which analysis of microbiome variability in health identified the primary parameters associated to microbiome composition. In this presentation\, I will discuss our experiences in large-scale microbiome monitoring\, show how the development of dedicated computational approaches can assist in microbiome analysis and interpretation\, and which confounders are essential for inclusion in microbiome disease research.\nIn addition\, I will show how Quantitative Microbiome Profiling (QMP; Vandeputte et al. Nature 2017)\, which combines microbiomics with flow cytometry-based cell counts\, is profoundly changing our view on gut microbiota variation and allowed the identification of an inflammation-associated\, cross-disease enterotype. Leads from such QMP-based profiling are enabling the development of microbiome modulation strategies.
