SUMMARY:In search of molecular Shangri-La: p16High senescence restricts cellular plasticity during somatic cell reprogramming
DESCRIPTION:Speaker: Prof. Dmitry Bulavin DR1 INSERM\, Institute for Research on Cancer and Aging (IRCAN)\, Nice\, France. \nDespite advances in four factor (4F)-induced reprogramming (4FR) in vitro and in vivo\, how it interconnects with senescence remains largely under-investigated. Using genetic and chemical approaches to manipulate senescent cells\, we found that removal of p16High cells resulted in 4FR of somatic cells into totipotent-like stem cells. These cells expressed markers of both pluripotency and the 2-cell (2C) embryonic state\, readily formed implantation-competent blastocyst-like structures\, blastoids\, and following morula aggregation\, contributed to embryonic and extraembryonic lineages in E12.5 embryos. We identified senescence-dependent regulation of nicotinamide N-methyltransferase (NNMT) as a key mechanism controlling the S-adenosyl-L-methionine (SAM) levels during 4FR that was required for expression of the 2C genes and acquisition of an extraembryonic potential. Importantly\, a partial 4F epigenetic reprogramming in old mice was able to significant reverse several histopathological markers of liver aging only in conjunction with depletion of p16High cells. \nOur results show that the presence of p16High senescent cells limit cell plasticity\, while their depletion can promote totipotency and tissue rejuvenation during 4F-reprogramming.

SOLTI is an academic group that promotes\, designs and develop clinical trials for cancer research\, especially for breast cancer. During the event\, they will talk about the origin and design of clinical trials using new biomarkers in the oncology field. Prospective biomarker validation is a critical and necessary step for its implementation in the clinical practice. Using real examples of clinical trials\, they will explain and discuss how a new biomarker\, initially validated with pre-clinical and/or translational data\, can be translated into a clinical study.

SUMMARY:Phys2BioMed Symposium
DESCRIPTION:The mechanical properties of biological samples may provide insights into many physiological and pathological processes. Examples are the mechanical properties of cancer cells or changes of mechanical properties during cell invasion. Defining standardized procedures for nanomechanical measurements and providing\, in the longer-term\, a data bank of mechanical fingerprints of diseases\, can lead to the development of effective early-diagnostic tools. \nPhys2BioMed is an ITN (Innovative Training Network) that has the purpose of providing scientific and technological outcomes on biomechanics and mechanical determinants of diseases. The key advantage is merging diverse competences at the European level\, from different fields like nanoscience and nanotechnology\, physics\, biology\, and medicine. Thanks to the participation of hospitals and companies\, ESRs (Early Stage Researchers) are exposed also to the non-academic sector. \nThe scientific community is invited to participate in person or online in this symposium. \nCheck for more information about the PHYS2BIOMED project here.

SUMMARY:From patterns to processes: towards a mechanistic understanding of the human microbiome
DESCRIPTION:Speaker: Stefany Moreno-Gámez \nPostdoc Fellow\, Massachusetts Institute of Technology (MIT) and the Broad Institute\, USA \nMost of our knowledge about the human microbiome comes from high-throughput metagenomic studies which have yielded large catalogues of bacterial species and genes that are correlated with health and disease. While useful\, these catalogues offer limited insights on how bacteria interact with one another and respond to their environments\, which is essential to understand the forces that shape community assembly and evolution in the human host. In this talk\, I will present our work on resource utilization strategies in the gut microbiome and show how characterizing the physiology and ecological interactions of individual microbes can help us understand the evolution of strategies for fiber degradation. In particular\, I will focus on the question of why strategies for private and public utilization of fibers coexist in the gut and will discuss the implications of this functional perspective for the development of fibers as prebiotics.

SUMMARY:Deciphering the molecular mechanism of human disease using network based methods
DESCRIPTION:Speaker: Iñigo Barrio Hernández \nPostdoc Fellow\, EMBL’s European Bioinformatics Institute (EMBL-EBI)\, Wellcome Genome Campus\, UK \nProteins that interact within molecular networks tend to have similar functions and when perturbed\, influence the same organismal traits. Using an interactome to propagate starting signals coming from different sources we can generate profiles to do trait to trait comparaisons. We can also detect communities of proteins tightly interacting within each other\, cores of cell signalling related with pathogenesis and likely hotspots of therapeutic targets. In this talk I will introduce two examples of the potential of network based methods to: establish a pleiotropy map of human disease using common genetic variation (GWAS) associated to more than 1000 traits\, and increase our knowledge of the role of astrocytes in a neurodegenerative disease (ALS)\, integrating different omics data.

SUMMARY:Targeting RNA modifications in cancer
DESCRIPTION:Speaker: Dr. Michaela Frye – Professor\, Mechanisms Regulating Gene Expression\, Deutsches Krebsforschungszentrum\, Heidelberg\, Germany. \nMany of the hundreds of known chemical modifications in RNA were discovered over forty years ago but then forgotten because suitable\, sensitive tools to detect the modifications at high resolution were lacking. Through the development of novel biochemical\, functional and genomics tools we are only now beginning to understand the whole breadth and extensive functional roles of RNA modifications in higher organisms. \nMichaela Frye will present some mechanistic examples how RNA modifications help to shape normal tissue homeostasis\, and how aberrant formation of RNA modifications contributes to human diseases such as cancer. She will focus on the functional roles of selected RNA modifications in regulating metabolic pathways and how they contributes to tumor metastasis and drug resistance. Together\, their work demonstrates that by understanding the role of RNA modifications in physiology and pathology\, novel therapeutic strategies for human diseases can be identified and further optimized for clinical studies.

SUMMARY:Repurposing the principles of embryonic development and reproductive biology as a potential solution for the diseases of aging
DESCRIPTION:Speaker: Dr. Vittorio Sebastiano – Associate Professor of Obstetrics and Gynecology (Reproductive and Stem Cell Biology)\, Stanford School of Medicine\, Stanford\, CA\, United States. \nAging is characterized by a gradual loss of function occurring at the molecular\, cellular\, tissue and systemic levels. At the chromatin level\, progressive accumulation of epigenetic errors eventually leads to a broad spectrum of outcomes including aberrant gene regulation\, stem cell exhaustion\, senescence\, and deregulated cell/tissue homeostasis. Notably\, while aging affects the single individuals of a given species\, it does not affect the species as a whole\, and no vertical transmission of parents’ age has ever been observed. \nHis hypothesis is that embryonic development and reproductive biology hold the secret to prevent or even reverse cellular aging and hence\, tissue and systemic aging. \nBased on this hypothesis\, the focus of the Sebastiano Lab is to study and model human embryonic/reproductive development\, dissect its principles\, and apply them to somatic cells as a potential solution to combat aging-associated diseases. \nThey utilize two complementary approaches. First\, through the cell autonomous rejuvenation of cells using a technology (Epigenetic Reprogramming of Aging\, ERA) that the lab has pioneered (Sarkar et al.\, 2020); second\, by in vitro engineering thymic and gonadal tissues as a potential solution to provide systemic enhanced health span to elderly individuals.
