DESCRIPTION:Dear all\, \nThe next IBEC Seminar will take place on Wednesday 05th October at 10:00\, when David Odde\, from Department of Biomedical Engineering at University of Minnesota\, will be talking about «Differential migration mechanics and immune response of glioblastoma subtypes». \nDavid Odde\, Department of Biomedical Engineering at University of Minnesota \nGlioblastoma remains a deadly cancer driven by invasion of tumor cells into the brain. Transcriptomic analyses have revealed distinct molecular subtypes\, but mechanistic and targetable differences that explain clinical differences are not clear. Using a state-of-the-art immunocompetent mouse model for glioblastoma – where tumors are induced by injection of plasmids containing human glioblastoma subtype-defining genetic drivers in a wild-type background – we found that\, as predicted by the motor-clutch model for cell migration (Klank et al.\, Cell Rep\, 2017)\, mesenchymal glioma cells are more spread\, generate larger traction forces\, and migrate faster in brain tissue compared to proneural cells. Despite their fast migration and comparable proliferation rate in vitro\, mice with mesenchymal tumors live longer than mice with proneural tumors\, which was correlated with an immune response in the mesenchymal mice that included T cell-mediated killing of cancer cells\, similar to human tumors. Thus\, mesenchymal tumors have aggressive migration\, but are relatively immunologically ‘hot’ which suppresses net proliferation\, features which are captured by our Brownian Dynamics tumor simulator (Klank et al.\, Conv Sci Phys Oncol\, 2018). These two features counteract each other and may explain the lack of a strong survival difference between subtypes clinically\, while also opening up new opportunities for subtype-specific therapies.

SUMMARY:Peptides and bioinstructive polymers on surfaces for prevention of oral infections
DESCRIPTION:Conrado Aparicio\, Faculty of Odontology\, UIC Barcelona and Associated Researcher – IBEC \nOral infections are the most prevalent infectious diseases worldwide. We have explored bioinspired modification of surfaces of restorative/regenerative materials and implants as well as dental tissues to address oral infections by harnessing the biomolecular toolbox –oligopeptides\, proteins\, recombinamers\, … and thus\, mitigating the worldwide threat of antimicrobial resistance. \nOne of our strategies aimed at exploiting secondary structure and self-assembly of anti-biofilm peptides and recombinamers to increase anti-biofilm potency vs relevant oral biofilms and to form highly hydrophobic interfaces to obtain dual-action surfaces to address infection in dental peri-tisular and peri-implant sites. \nAlternatively\, we have found inspiration in the natural junction at the tooth-oral mucosa interface. Teeth\, long-lasting percutaneous organs\, feature soft tissue attachment through adhesive structures\, hemidesmosomes\, in the junctional epithelium basement membrane adjacent to teeth. This soft tissue attachment prevents bacterial infection of the tooth despite the rich – and harsh – microbial composition of the oral cavity. Consequently\, we have tapped basement membrane peptides known to engage specific integrins as well as synthesized new bioinstructive photocurable resins that stimulate formation of hemidesmosomes on synthetic and natural surfaces. \nWe recently have also combined those strategies with immunomodulatory regulation to reduce inflammation triggered by pathogenic biofilms causing oral infections.

SUMMARY:Enhancement of anti-tumor immunity in RAS-addicted cancers with tri-complex RAS(ON) inhibitors
DESCRIPTION:Speaker: Elsa Quintana\, Pharm.D.\, Ph.D. Senior Director Cancer Immunology\, Revolution Medicines\, Redwood City\, USA \nRAS mutations (RASMUT)\, the most frequent oncogenic alterations in human cancers\, promote carcinogenesis through sustained cellular proliferation and also shape the tumor microenvironment to allow cancer cells to evade an anti-tumor immune response. \nRAS oncoproteins may be targeted with mutant selective\, tri-complex RAS(ON) inhibitors or tri-complex RASMULTI(ON) inhibitors. These inhibitors have distinct preclinical profiles and potential clinical applications. KRASG12C(ON) mutant-selective inhibitors potently inhibit the RAS pathway in KRASG12C mutant cancer cells and indirectly modulate the TME by impacting the secretion of cytokines and the expression of cancer cell surface proteins. A RASMULTI(ON) inhibitor targets not only multiple RAS mutants but also wild-type RAS\, a critical signaling node both in tumors and normal immune cells. We have evaluated the effects of KRASG12C(ON) and RASMULTI(ON) inhibitors on the TME and anti-tumor immunity in murine tumor models in vivo. \nOral daily administration of either a KRASG12C(ON) inhibitor or a RASMULTI(ON) inhibitor drove complete tumor regressions and synergized with anti-PD-1 in vivo in an immune-sensitive colorectal KRASG12C mutant model. Re-challenge experiments demonstrated that both inhibitor classes induce immunological memory. RAS inhibition with mutant-selective or RASMULTI(ON) inhibitors decreased the expression of checkpoint molecules\, stem cell markers and proliferation markers on KRASG12C mutant tumor cells in vitro and in vivo. Single-agent therapy induced an increase of CD8+ T-cells and dendritic cells (DCs) and a decrease in pro-tumorigenic M2 macrophages and myeloid derived suppressor cells (MDSCs) in the tumor. In tumors treated with KRASG12C(ON) inhibitors this effect on immune cell infiltration was\, in part\, mediated through the modulation of tumor secreted cytokines. A RASMULTI(ON) inhibitor induced a more pronounced decrease in M2 macrophages which raised the question whether\, in addition to the impact on cytokine production\, there was also a direct effect of RAS inhibition on macrophages. A RASMULTI(ON) inhibitor\, similar to SHP2 inhibitors (Quintana\, E. et al\, 2020)\, but unlike a mutant selective RAS(ON) inhibitor\, directly and selectively reduced the viability of M2\, but not M1 macrophages in vitro. To interrogate if these properties can be leveraged to convert the TME of an immune-suppressive tumor we characterized the impact of RAS(ON) inhibitors in an anti-PD-1-refractory lung KRASG12C mutant model; we observed a significant conversion of the TME in favor of anti-tumor immunity in this model. \nThese preclinical data suggest that mutant-selective and\, more profoundly\, RASMULTI(ON) inhibitors promote the conversion of an immune-suppressive into an ‘immune-hot’ TME in RAS-mutant tumors.

SUMMARY:Regulation of hematopoietic stem cell dormancy
DESCRIPTION:Speaker: Nina Cabezas-Wallscheid\, Ph.D. Group Leader\, Regulation of Hematopoietic Stem Cell Dormancy\, Max Planck Institute\, Freiburg\, Germany. \nBone marrow hematopoietic stem cells (HSCs) are vital for the maintenance of hematopoiesis. In inbred mice housed in gnotobiotic facilities\, the top of the hematopoietic hierarchy is occupied by dormant HSCs\, which reversibly exit quiescence during stress. Still\, whether HSC dormancy exists in human remains unclear. Our recent work shows the existence and relevance of dormancy in human HSCs and identifies the hyaluronic acid-GPRC5C signalling axis as an essential component controlling the HSC state (Zhang et al.\, Nature Cell Biology 2022). Further\, HSCs rely on complex regulatory networks to preserve their function. Due to the scarcity of HSCs\, technical challenges have limited our insights into the interplay between HSC metabolism and their transcriptional and epigenetic regulation. We recently established and integrated low-input multi-layer OMICs data revealing distinct metabolic and epigenetic hubs that are enriched in HSCs and their downstream multipotent progenitors (Schönberger et al.\, Cell Stem Cell 2022). Mechanistically\, we uncover a non-classical retinoic acid signaling axis that regulates HSC identity. Our findings emphasize how a single metabolite controls stem cell fate by instructing epigenetic and transcriptional attributes.

SUMMARY:The roles of mobile DNA in human cancer
DESCRIPTION:Speaker: Jose Tubio\, Ph.D. Group Leader\, Mobile Genomes lab\, Centre for Research in Molecular Medicine and Chronic Diseases (CIMUS)\, University of Santiago de Compostela\, Spain. \nSomatic retrotransposition represents the most frequent class of structural variation in some human cancers. For the last 10 years\, we have illuminated two relevant mutational mechanisms by which somatic RT can provide the mutations that a cancer clone requires to survive and grow (PMID: 25082706; PMID: 32024998). Currently\, our research focuses on the use of long-read sequencing and chromatin conformation capture techniques for the discovery of novel\, previously unreported mutational roles of somatic retrotransposition in human cancer. \nJose Tubio has carried out his research at the forefront of cancer genomics\, being involved in the discovery of new cancer genes in hematological cancers and bone cancers\, the identification of new mechanisms of mutation\, the characterization of mechanisms that drive the dynamics of metastasis\, the identification of genes that drive transmissible cancers\, and the identification of early genetic mutations that occur in normal tissues and their role in tumour transformation.
