SUMMARY:Aberrant phase separation is a widespread killing mechanism of bioactive peptides in health and diseas
DESCRIPTION:Steven Boeynaems\, Assistant Professor at Baylor College of Medicine and Investigator at the Texas Children’s Hospital \nCationic repeat peptides are increasingly implicated in the pathophysiology of age-related neurodegenerative disease. These peptides are highly toxic in all tested disease models\, and seem to perturb an ever-expanding list of cellular pathways. This observation begs the question whether this target promiscuity is (1) relevant to disease and (2) how it is encoded in such simple sequences. A first clue came from the observation that these cationic repeat peptides have the tendency to drive protein condensation. Given their charge\, these peptides are exquisitely sensitive to the presence of nucleic acids. By studying their target space as a function of RNA concentration using mass spec\, we uncover a hidden hierarchy in their target preference that relates to subcellular differences in RNA abundance. \nTo test whether these insights from disease translate to biology\, we used this framework to develop a machine learning algorithm that allowed us to predict protein behavior form sequence. Compellingly\, we uncover that bioactive peptides such as antimicrobial peptides produced by our own cells\, but also venom peptides derived from spiders\, scorpions and rattle snakes\, phenocopy the molecular behavior of these disease-related cationic peptides. In all\, we find that such peptides exert toxicity by upsetting the electrostatic balance of a cell\, hitting myriad pathways at once. We name this process polycation poisoning and find that it has independently evolved again and again across the entire tree of life as a killing strategy of bioactive peptides. Based on these findings we are now exploring toxin-antitoxin systems as innovative therapeutic strategies to protect neurons against cationic repeat peptide toxicity. \nHosts: Benedetta Bolognesi and Xavier Salvatella

SUMMARY:Bake Sale per col·laborar amb Babies Uganda
No t’ho perdis!

SUMMARY:Metabolism in the single cell era
DESCRIPTION:Speaker: Dr. Rafael J. Argüello \nPostdoctoral Researcher \nCNRS Researcher / ISAC Marylou Ingram Scholar 2021-2023 / “Metabolic Regulation of Immunity” Centre d`Immunologie de Marseille-Luminy\, France. \nMetabolic reprogramming is a hallmark of cancer and immune responses. Different immune cells can adopt pro or anti-tumoral functions that are correlated with the dependence of different energetic metabolism pathways. Metabolic reprogramming involves dramatic changes in cellular levels of NAD+\, Lactate\, Acetyl-CoA\, SAM and other metabolites that are substrates of the enzymes that mediate epigenetic modifications.  During immune activation epigenetic changes of the chromatin occur concomitantly to changes in the metabolism. These links between metabolic changes in cancer and immune cells\, their impact on epigenetics\, gene expression and cell function are poorly understood. Especially due to the lack of adapted technologies that allow to study metabolism and epigenetics with single cell resolution in physiologically relevant contexts.\nIn this seminar I will present our recent development and applications of a method to monitor functional metabolism with single cell resolution\, and our first results using this technology\, called SCENITH in combination with measurements of epigenetic marks of the chromatin (Epic-SCENITH). Altogether\, SCENITH enable to characterize the metabolic and epigenetic landscapes of cancer and immune cells in patient samples. We envision to use SCENITH as a personalized medicine approach and use its information to predict the efficacy of immunotherapies.

SUMMARY:Novel transcriptional regulation and therapeutic targets in T-Cell Leukemia
DESCRIPTION:Speaker: Daniel Herranz\, Pharm. D. Ph.D. Assistant Professor of Pharmacology – Cancer Pharmacology Liaison for Center of Outreach and Engagement – Robert Wood Johnson Medical School – Rutgers\, Cancer Institute of New Jersey –  Rutgers\, The State University of New Jersey – US \nAbstract: \nT-ALL is a NOTCH1-driven hematological malignancy in which 20% of the patients lose the expression of the tumor suppressor PTEN. Still\, not all of these cases can be explained by prototypical mutations/deletions in its coding sequence\, which led us to hypothesize there might be additional mechanisms controlling PTEN expression. Indeed\, using comprehensive epigenetic profiling\, chromosome conformation techniques and novel mouse models\, we identified a novel enhancer regulatory region of PTEN (PE) that regulates its expression in T-cells and is recurrently deleted in T-ALL patients\, providing a new mechanism for the loss of PTEN expression. \nIn addition\, using a variety of mouse models\, epigenetic profiling and unbiased acetyl-proteomics\, we have identified a novel therapeutically targetable NOTCH1-SIRT1-KAT7 axis in T-ALL. Sirt1 loss in T-ALL in vivo shows highly antileukemic and synergistic effects with NOTCH1 inhibition. Mechanistically\, our data shows that NOTCH1 directly regulates SIRT1 through an enhancer region (N-Se)\, and SIRT1 itself deacetylates and activates the histone acetyltransferase KAT7. Consequently\, hyperacetylated KAT7 upon SIRT1 loss is less active\, and this partially mediates the antileukemic effects of SIRT1 loss.
