SUMMARY:Targeting KRAS: Light at the End of the Tunnel
DESCRIPTION:Speaker: Mariano Barbacid\, PhD – AXA-CNIO Professor of Molecular Oncology – Centro Nacional de Investigaciones Oncológicas (CNIO) – Spanish National Cancer Research Center – Madrid\, Spain. \nKRAS\, an oncogene responsible for one fourth of all human cancers\, was discovered four decades ago. Yet\, the first selective inhibitor was not approved until 2021. In spite of this long-awaited breakthrough\, current KRAS inhibitors only block one of the multiple oncogenic isoforms\, KRASG12C which is primarily present in lung adenocarcinomas. Moreover\, tumor resistance to KRAS inhibitors appears to be a significant issue in the clinic (Awad et al.\, N Engl J Med\, 2021; Zhao et al.\, Nature 2021). In an effort to provide experimental information that might help to improve current KRAS targeting strategies\, we have generated two genetically engineered mouse models of Kras/Trp53-driven lung adenocarcinoma to (i) interrogate the mechanisms of drug resistance that appear in KRASG12C driven tumors and (ii) compare the therapeutic consequences of eliminating instead of inhibiting KRAS oncoproteins. Our results indicate that Kras gene amplification along with activation of drug metabolic pathways are major components of the mechanisms responsible for the acquisition of resistance. More importantly\, genetic evidence strongly supports the concept that Kras ablation provides superior therapeutic benefits than Kras inhibition\, thus arguing for the development of Kras degron strategies.

DESCRIPTION:Speaker: Jose Ordovas-Montanes\, PhD – GL – Research Faculty – Division of Gastroenterology – Boston Children’s Hospital – Assistant Professor of Pediatrics – Harvard Medical School – Robertson Investigator – New York Stem Cell Foundation – Principal Faculty – Harvard Stem Cell Institute. \nFor a barrier tissue to effectively learn from previous immunological experiences\, it can sense\, adapt\, and store this information in essential or resident cell types. The foundational architecture of barriers includes essential stromal and stem-cell derived epithelial cell subsets that collectively perform key tissue functions. These cell subsets are critical for responding to\, and learning from\, environmental challenges\, together with the support of resident immune cells. However\, the ability of barrier tissues such as the skin\, gut or airway to efficiently adapt can become dysregulated during acute infection with viruses (SARS-CoV-2) or chronic disease states (inflammatory bowel disease). Identifying the essential cell types\, states and circuits of communication which predict and sustain disease in humans is vital to effectively treating these conditions. Our work across barrier tissues suggests that deriving insights from single-cell measurements can identify predictive determinants of organismal health and disease.

SUMMARY:Deep mutational scanning of entire human protein interaction domain families
DESCRIPTION:Protein interaction networks represent the backbone of the cell’s functional organization. Their architecture relies on the specificity of these interactions. Mutations that alter specificity may rewire the network and the way it corrdinates biological processes\, and lead to diseases or functional innovations. Understanding how the sequence of a protein determines the specificity of its interaction profile thus represents an important step towards a model of the genotype-phenotype map. To adress the sequence-function relationship`in protein-protein interactions\, we conducted a deep mutational scanning of the entire human basic leucine zipper (bZIP) interaction network. This interaction domain family\, composed of 54 members in humans\, has served as a model system for the study of protein interaction specificity. While the general rules of bZIP specificity are relatively well known\, it remains impossible to predict interaction partners accurately. Using our established deepPCA system\, we measured the effect of all single amino acid substitutions in all 54 members\, on the interaction with all 54 wild-type members. This dataset of ~2M quantitative protein-protein interaction measurements will enable training a deep neural network that captures the sequence-function relationship in order to quantitatively predict interaction between bZIPs from sequence alone. I will present the dataset and the deep learning model\, as well as preliminary data on two other families\, basic helix-loop-helix and SH3 domains\, for which we plan to collect 6 and 20M data points\, respectively. We foresee that applying this strategy on a variety of domain families of different architecture will enable the development of general models that can predict the affinity of an interaction from the sequenece of the two partners.

La formación estará impartida por Estibaliz Urarte\, comunicadora científica\, y se enmarca dentro de las actividades del programa “Recerca en Societat” que tiene por objetivo fomentar vocaciones científicas y mejorar la difusión de la ciencia. \nEstibaliz Urarte es bióloga de formación y ha estado 8 años vinculada al mundo de la investigación en el área de la Fisiología y Bioquímica Vegetal. Posteriormente orientó su carrera hacia el ámbito de la comunicación científica y la divulgación. Se formó en el área de la SciComm cursando el posgrado en Comunicación Científica de la UVic y en la actualidad trabaja en dicho ámbito en el Área de Participación del Paciente en la Investigación de la Fundació de Recerca Sant Joan de Déu. Paralelamente\, escribe sobre ciencia para una revista cultural de carácter local y realiza colaboraciones puntuales con blogs y revistas de ciencia\, como Mujeres con Ciencia y Principia Magazine. También es secretaria general de la Asociación Catalana de Comunicación Científica y coordinadora del Grupo de Perspectiva de Género de dicha asociación. Desde 2021 es profesora de Divulgación Científica y Difusión de la I+D+i en la UIC dentro del grado de Ciencias Biomédicas.

SUMMARY:Proteostasis of Aging and Stem Cells
DESCRIPTION:Speaker: David Vilchez\, PhD – Professor\, CECAD Cluster of Excellence: Cellular Stress Responses in Aging-Associated Diseases\, Medical Faculty\, University of Cologne\, Germany. \n«While it has long been noted that genome stability is a central function required for survival of stem cells\, the role of proteostasis has not been explored. With the asymmetric divisions invoked by stem cells\, the passage of damaged proteins to daughter cells can destroy the resulting lineage of cells and possibly accelerate the aging process. Furthermore\, the retention of damaged proteins by the stem cell could result in diminished stem cell function and premature aging. Therefore\, a firm understanding of how stem cells maintain their proteostasis is of central importance. Because experiments with mammalian embryonic stem cells have clearly demonstrated their capacity to replicate continuously in the absence of senescence\, we hypothesize that these cells could provide a novel paradigm to study the regulation of proteostasis and its demise in aging. We have recently described that human embryonic stem cells (hESCs) exhibit high proteasome activity compared to their differentiated counterparts. This enhanced proteasome activity is necessary for hESC function. Furthermore\, we have uncovered that PSMD11/rpn-6\, a key proteasomal subunit\, is required for this activity and its mode of regulation and conservation in the aging process of the invertebrate C. elegans. Our findings established RPN-6 as a potent regulator of proteasome activity that alleviates the deleterious effects associated with aberrant protein aggregation\, providing a powerful candidate to correct proteostatic deficiencies in disorders such as HD. Moreover\, our results in C. elegans led us to find that FOXO4\, a transcription factor associated with longevity\, regulates proteasome activity in hESCs and is necessary for hESCs differentiation into neural cells. Therefore\, our results established a novel regulation of proteostasis in hESCs that links longevity and stress resistance in invertebrates with hESC function and identity».

Therefore\, we organize this seminar to help Ukrainian refugees to get familiar with the way CV is prepared in Spain\, the core information to be included and omitted and different tips of how to look for jobs in the country. Knowing all the language difficulties\, the seminar is going to be translated in Ukrainian.

SUMMARY:IBMB Seminars: Temporal Crontol of Neurogenesis
DESCRIPTION:Speaker: Bassem Hassan\, PhD. \nScientific Director and Deputy General Director at the\nInstitut du Cerveau – Paris Brain Institute (PBI). \nAn important manifestation of time during neurogenesis is the generation of different types of neurons from the same pool of progenitors over time. This means that neural progenitors can decode time and convert it into discrete quanta of different neuronal fates. However\, for progenitors to decode time into discrete quanta\, it must be true that total time is somehow genetically encoded. The extension and retraction of this total neurogenic time in closely related species is associated with changes in brain size\, often without significant changes to neuronal fate. However\, the genetic mechanisms that translate total time into neuronal numbers are not well understood. Previous work from the lab suggests that transcription factors known as proneural proteins are good candidates for this link: altering the timing of proneural protein expression or activity alters the size of the neuronal lineages they produce. Conversely\, changes the total time of neurogenesis alters proneural gene expression. These observations suggest an intimate mechanistic link between timing\, transcriptional regulation and brain size.

SUMMARY:Shooting with electrons to unravel mechanistic insights of the human neutral amino acid transporter ASCT2 (SLC1A5)
DESCRIPTION:Speaker: Dr. Cristina Paulino \nAssociate Professor and Head of the cryo-Electron Microscopy Unit \nDepartment of Structural Biology and Membrane Enzymology – Groningen Biomolecular Sciences and Biotechnology Institute (GBB) \nUniversity of Groningen\, The Netherlands \nMy research aims at elucidating the mechanism of action of membrane transporters on a molecular level. To address these questions\, we use an interdisciplinary approach with cryo-EM as a central technique and contrast findings with comprehensive functional and computational studies. My talk will highlight how several advantages of cryo-EM can be exploited to study these pharmacological relevant transporters\, taking our studies on ASCT2 as an example. ASCT2 belongs to the SLC1A superfamily and is the main source of glutamine uptake in human cells. Its upregulation is strongly linked to cancer cell growth and poor patient survival\, making it a potential target in cancer therapy. We were able to solve the first structures of ASCT2\, and answer key mechanistic questions. Our observations are of great fundamental interest and pave the way for the rational design of inhibitors.

SUMMARY:Unraveling the complexity of cellular senescence
DESCRIPTION:Speaker: Federico Pietrocola\, PhD – GL – Department of Biosciences and Nutrition – Karolinska Institutet – Sweden. \nThe role of senescent cells in aging and age-related pathologies is well established. Nonetheless\, senescent cells exhibit remarkable – functional and molecular – heterogeneity\, which limits the implementation of senescence-targeted therapies. In the lab\, we are specifically interested in resolving the complexity of the senescent phenotype\, aiming to shed light on the vulnerabilities that characterise pathologically meaningful senescent cells. Our current work focuses on identifying metabolic addictions of senescent cells\, with particular emphasis on the role of arginine metabolism in the establishment and maintenance of the senescent state.
