SUMMARY:Overcoming multidrug resistance in cancer with dirty drugs
DESCRIPTION:Resistance to therapies\, intrinsic or acquired during treatment\, is a major medical problem that limits the efficacy of our current anti-cancer armamentarium. Unfortunately\, while some of the mechanisms that contribute to resistance are known\, our knowledge of this phenomenon\, particularly for the case of multi-drug resistance (MDR)\, remains incomplete. Through genome-wide genetic screens and bioinformatic analyses\, we discovered that one of the most frequent mutations in cancer\, FBXW7 deficiency\, confers a very severe MDR phenotype to cancer cells. Interestingly\, we later found that these MDR is associated to an increase in mitochondrial activity\, which was previously linked to resistance by other groups. While we do not understand how mitochondrial function relates to MDR\, it offered us a vulnerability that can be exploited. Accordingly\, therapies that target mitochondria are particularly toxic for FBXW7 deficient cells. Finally\, when looking for additional drugs that could overcome the MDR of FBXW7 mutant cells through bioinformatics\, we discovered a group of seeming unrelated multi-kinase inhibitors that were able to do it. Interestingly\, these compounds are “dirty” drugs that are often used in the clinic\, even though their actual target is unknown. We believe we have discovered an alternative model that explains why these drugs kill cancer cells. But if you want to know about it\, you will have to come to my seminar.

SUMMARY:Galectins: emerging glyco-checkpoints linking immune and vascular programs in inflammation and cancer
DESCRIPTION:The responsibility for deciphering the biological information encoded by the glycome- the whole repertoire of glycans in cells and tissues- is assigned to endogenous glycan-binding proteins or lectins whose expression is regulated at sites of inflammation and tumor growth. With the goal of identifying novel therapeutic targets in cancer and autoimmune diseases\, our laboratory investigates the molecular interactions between glycosylated receptors and glycan-binding proteins\, particularly galectins in the control of immune and vascular programs. In the past years\, we have identified essential roles for galectin-1\, a proto-type member of the galectin family\, in promoting immune evasion in different tumor models and limiting autoimmune inflammatory responses by selectively modulating effector and regulatory T cells and instructing the differentiation of tolerogenic dendritic cells and anti-inflammatory macrophages\, Moreover\, we identified the contribution of glycosylation-dependent\, galectin-driven circuits to  mechanisms of resistance of anti-angiogenic and immunotherapeutic modalities. More recently we validated the relevance of galectin-1-glycan programs in models of neurodegenerative and cardiovascular diseases. Thus\, galectins may function as regulatory checkpoints that translate ‘sugar codes’ into immune and vascular signaling programs in tumor and inflammatory microenvironments\, highlighting their therapeutic potential in a broad range of pathologic conditions.

SUMMARY:High-throughput functional characterization of phosphorylation sites in yeast
DESCRIPTION:Phosphorylation is a critical post-translational modification involved in the regulation of almost all cellular processes. However\, fewer than 5% of thousands of recently discovered phosphosites have been functionally annotated. In this study\, we devised a chemical genetic approach to study the functional relevance of phosphosites in Saccharomyces cerevisiae. We generated 474 yeast strains with mutations in specific phosphosites that were screened for fitness in 102 conditions\, along with a gene deletion library. Of these phosphosites\, 42% exhibited growth phenotypes\, suggesting that these are more likely functional. We inferred their function based on the similarity of their growth profiles with that of gene deletions and validated a subset by thermal proteome profiling and lipidomics. A high fraction exhibited phenotypes not seen in the corresponding gene deletion\, suggestive of a gain-of-function effect. For phosphosites conserved in humans\, the severity of the yeast phenotypes is indicative of their human functional relevance. This high-throughput approach allows for functionally characterizing individual phosphosites at scale.
