Este estudio supone una estrategia prometedora para el desarrollo de nuevos tejidos cut\u00e1neos obtenidos con bioingenier\u00eda, f\u00e1cilmente reproducibles y rentables econ\u00f3micamente, un avance particularmente beneficioso en ensayos precl\u00ednicos de productos cosm\u00e9ticos y farmac\u00e9uticos, y abre camino a la creaci\u00f3n de complejos modelos in vitro de tejidos humanos.<\/strong><\/p>\n<p>La demanda de tejidos cut\u00e1neos sintetizados en el laboratorio ha aumentado mucho en los \u00faltimos a\u00f1os por sus aplicaciones en entornos cl\u00ednicos, como la sustituci\u00f3n de piel y los ap\u00f3sitos avanzados para heridas, as\u00ed como en escenarios precl\u00ednicos para testar nuevos f\u00e1rmacos y cosm\u00e9ticos y desarrollar modelos de enfermedades.<\/p>\n<p>Un tejido de piel sintetizado mediante bioingenier\u00eda debe ser f\u00e1cil de preparar, resistente mec\u00e1nicamente y propicio para la adhesi\u00f3n y el crecimiento celular, tanto en el compartimento epid\u00e9rmico como en el d\u00e9rmico. Debe lograr un equilibrio entre la elasticidad y la estabilidad mec\u00e1nica, y a la vez, proporcionar un microambiente celular adecuado para contener los diferentes compartimentos de la piel humana. Y uno de los puntos clave del proceso la correcta elecci\u00f3n del material del \u201candamio\u201d sobre el que se construir\u00e1 el tejido.<\/p>\n<p>Recientemente, un equipo de cient\u00edficas liderado por <strong>Elena Mart\u00ednez, <\/strong>investigadora principal\u00a0del grupo\u00a0<a href=\"https:\/\/ibecbarcelona.eu\/es\/biomsyscelleng\" target=\"_blank\" rel=\"noopener\">Sistemas Biomim\u00e9ticos para Ingenier\u00eda Celular<\/a> del IBEC, han publicado en la revista International Journal of Bioprinting un trabajo que ayudar\u00e1 a abordar y resolver estas cuestiones. En concreto, presentaron una\u00a0biotinta entrecruzable\u00a0mediante luz l\u00e1ser, dise\u00f1ada para sintetizar piel humana, basada en formulaciones de tiol-norborneno-pululano (N-PLN) combinadas con varios fotopolimerizadores.<\/p>\n<p>Esta nueva biotinta se utiliz\u00f3 de manera efectiva en una bioimpresora de escritura por l\u00e1ser desarrollada en el laboratorio, para generar matrices d\u00e9rmicos epitelializadas. El compartimento d\u00e9rmico se form\u00f3 mediante la fotopolimerizaci\u00f3n de una soluci\u00f3n de prepol\u00edmero que conten\u00eda fibroblastos humanos, mientras que el compartimento epid\u00e9rmico se desarroll\u00f3 sembrando queratinocitos humanos en los hidrogeles previamente generados con fibroblastos embebidos. Utilizando luz visible, se pudieron imprimir hidrogeles de 2,5 mm\u00b3 con c\u00e9lulas en su interior, en solo 10 segundos. La qu\u00edmica de fotopolimerizaci\u00f3n mediante formulaciones de tiol empleada en este estudio cre\u00f3 una matriz extracelular bien definida con reticulaciones ortogonales, manteniendo altas tasas de viabilidad celular para fibroblastos encapsulados.<\/p>\n<p>Este estudio supone una estrategia prometedora para desarrollar un modelo de piel humana utilizando un sistema de bioimpresi\u00f3n 3D basado en luz hecho a medida y usando nuevos pol\u00edmeros N-PLN. Estos hidrogeles, formados despu\u00e9s de una breve exposici\u00f3n a la luz visible a baja dosis, exhiben las propiedades fisicoqu\u00edmicas necesarias para proporcionar una excelente viabilidad, proliferaci\u00f3n y elongaci\u00f3n celular, as\u00ed como la secreci\u00f3n de prote\u00ednas de matriz extracelular, cruciales para la correcta formaci\u00f3n de los tejidos.
