{"id":38633,"date":"2016-08-02T00:00:00","date_gmt":"2016-08-01T23:00:00","guid":{"rendered":"http:\/\/www.pcb.ub.edu\/\/dos-proteinas-preservan-las-celulas-madre-de-la-piel\/"},"modified":"2020-11-16T16:20:51","modified_gmt":"2020-11-16T15:20:51","slug":"dos-proteinas-preservan-las-celulas-madre-de-la-piel","status":"publish","type":"post","link":"https:\/\/www.pcb.ub.edu\/es\/dos-proteinas-preservan-las-celulas-madre-de-la-piel\/","title":{"rendered":"Dos prote\u00ednas preservan las c\u00e9lulas madre de la piel"},"content":{"rendered":"<p>Las prote\u00ednas identificadas,\u00a0Dnmt3a y Dnmt3b,\u00a0son esenciales para la auto-renovaci\u00f3n de las c\u00e9lulas madre de la piel\u00a0ya que con su acci\u00f3n se da el primer paso para que se dispare el programa gen\u00e9tico de c\u00e9lula madre.\u00a0\u201cSin ellas, esto no ocurre y las c\u00e9lulas madre se colapsan, desaparecen del tejido\u201d,\u00a0explica\u00a0Salvador Aznar Benitah, jefe del laboratorio\u00a0<a href=\"http:\/\/www.irbbarcelona.org\/en\/research\/stem-cells-and-cancer\" target=\"_blank\" rel=\"noopener noreferrer\">C\u00e9lulas Madre y C\u00e1ncer<\/a>\u00a0del IRB Barcelona.<\/p>\n<p>Lorenzo Rinaldi, estudiante de doctorado \u201cla Caixa\u201d y primer autor del trabajo, ha localizado cada una de las zonas del genoma donde se encuentran las prote\u00ednas. Rinaldi ha visto que, entre otras regiones,ambas prote\u00ednas act\u00faan sobre amplificadores gen\u00e9ticos (enhancers\u00a0y\u00a0superenhancers\u00a0en ingl\u00e9s), zonas alejadas de los genes que pueden aumentar hasta 200 veces la velocidad a la que se transcribe un gen.<\/p>\n<p>\u201cFue muy sorprendente ver que dos prote\u00ednas que siempre se han asociado a metilaci\u00f3n de ADN para reprimir genes, est\u00e9n encendiendo las zonas m\u00e1s activas de transcripci\u00f3n de genes de c\u00e9lula madre.Nunca las hab\u00edamos visto haciendo esta funci\u00f3n porque no se pod\u00edan investigar a nivel gen\u00f3mico las localizaciones globales de Dnmt3a y Dnmt3b. Gracias al avance de las t\u00e9cnicas de secuenciaci\u00f3n, m\u00e1s investigadores est\u00e1n viendo este mismo mecanismo que hemos descrito\u201d, explica\u00a0Rinaldi.<\/p>\n<p>De los 12.000 amplificadores gen\u00e9ticos que hay\u00a0en el genoma, alrededor de 300 sonsuperamplificadores gen\u00e9ticos relacionados con c\u00e9lulas madre. En todos ellos act\u00faan ambas prote\u00ednas para encender alrededor de los 1.000 genes necesarios para la auto-renovaci\u00f3n de las c\u00e9lulas madre. Mediante la metilaci\u00f3n del\u00a0superamplificador, activan esta primera pieza de la maquinaria que promover\u00e1 la expresi\u00f3n amplificada de estos genes esenciales para la c\u00e9lula madre.<\/p>\n<p>Asociaci\u00f3n con c\u00e1ncer<\/p>\n<p>Las c\u00e9lulas tumorales sufren muchos\u00a0cambios en la metilaci\u00f3n del ADN. Adem\u00e1s, en los \u00faltimos a\u00f1os, gracias a la secuenciaci\u00f3n masiva de los genomas de c\u00e9lulas tumorales se est\u00e1n identificando muchas\u00a0mutaciones en los amplificadores gen\u00e9ticos, no solo en los cuerpos de los genes. Finalmente, estas dos prote\u00ednas,\u00a0Dnmt3a y Dnmt3b, est\u00e1n alteradas en muchos tipos de tumores, como en leucemias, pulm\u00f3n y col\u00f3n.<\/p>\n<p>\u201cEstos tres elementos por separado est\u00e1n asociados al desarrollo de diferentes tipos de c\u00e1ncer.\u00a0Dado que estas prote\u00ednas activan los amplificadores de expresi\u00f3n g\u00e9nica a trav\u00e9s de la metilaci\u00f3n del ADN, creemos que ser\u00eda interesante estudiarlas en c\u00e9lulas cancer\u00edgenas para ver si tienen alguna funci\u00f3n en el desarrollo tumoral\u201d, concluye el Dr. Aznar-Benitah.<\/p>\n<p>El trabajo ha recibido financiaci\u00f3n del Ministerio de Econom\u00eda y Competitividad y de fondos FEDER.
