El trabajo, publicado en la revista <em>Frontiers<\/em>, pone de manifiesto el MYC como un motor central de la progresi\u00f3n agresiva de la LLC. Al mapear la actividad de MYC entre pacientes y etapas de la enfermedad, la investigaci\u00f3n ofrece nuevas perspectivas sobre qu\u00e9 pacientes pueden tener un riesgo m\u00e1s alto de progresi\u00f3n r\u00e1pida y apunta hacia posibilidades estrat\u00e9gicas de tratamiento personalizado. Este trabajo tambi\u00e9n contribuye a nuestra comprensi\u00f3n de la biolog\u00eda de la leucemia y proporciona una base para futuros desarrollos terap\u00e9uticos dirigidos a MYC.\u00a0<\/strong><\/p>\n<p>El estudio ha sido liderado por el <a href=\"https:\/\/www.cnag.eu\/teams\/genome-research-unit\/biomedical-genomics-group\" target=\"_blank\" rel=\"noopener\">grupo de Gen\u00f3mica Biom\u00e9dica<\/a> del CNAG, bajo la direcci\u00f3n de <strong>Ivo Gut<\/strong>, con la contribuci\u00f3n de la investigadora <strong>Maria Tsagiopoulou<\/strong>, que llev\u00f3 a cabo parte de esta investigaci\u00f3n mientras a\u00fan trabajaba en el CNAG. El estudio se ha centrado en c\u00f3mo la activaci\u00f3n de los genes controlados por MYC afecta el comportamiento del tumor y los resultados cl\u00ednicos.<\/p>\n<p>La leucemia linfoc\u00edtica cr\u00f3nica es un tipo de c\u00e1ncer que afecta a las c\u00e9lulas B maduras, un componente esencial de nuestro sistema inmunitario. En algunos pacientes, la enfermedad progresa lentamente y se mantiene estable durante a\u00f1os, mientras que en otros se vuelve agresiva. Un desarrollo especialmente grave es la transformaci\u00f3n de Richter, en la que este tumor evoluciona hacia un linfoma de crecimiento r\u00e1pido. Entender los factores moleculares que se encuentran detr\u00e1s de estos diferentes cursos de la enfermedad es crucial para mejorar la atenci\u00f3n a los pacientes y orientar los tratamientos.<\/p>\n<p>Para explorarlo, los investigadores analizaron cientos de muestras de pacientes procedentes de proyectos p\u00fablicos. Esto incluy\u00f3 secuenciaci\u00f3n masiva de ARN (<em>bulk RNA sequencing<\/em>), que mide la actividad global de los genes en muestras tumorales, y muestras emparejadas tomadas de los mismos pacientes antes y despu\u00e9s de la transformaci\u00f3n de Richter, lo que permite observar directamente los cambios a medida que la enfermedad progresa. Tambi\u00e9n estudiaron m\u00e1s de 40.000 c\u00e9lulas individuales mediante secuenciaci\u00f3n de ARN de c\u00e9lula \u00fanica (<em>single-cell RNA sequencing<\/em>), capturando tanto c\u00e9lulas tumorales como c\u00e9lulas inmunitarias del entorno. Combinando estos conjuntos de datos, el equipo cre\u00f3 una firma de 285 genes diana de MYC, ofreciendo una visi\u00f3n detallada de la actividad de MYC en diferentes etapas de la enfermedad.<\/p>\n<p>Los resultados mostraron que la actividad de MYC es m\u00e1s alta en los casos de leucemia linfoc\u00edtica cr\u00f3nica con caracter\u00edsticas m\u00e1s agresivas y aumenta a\u00fan m\u00e1s durante la transformaci\u00f3n de Richter.
