In total, it is estimated that 1.2 million people die each year due to this disease, which represents almost 5% of all annual deaths worldwide. Despite the enormous financial and<\/span><span data-contrast=\"auto\">\u00a0personal burden that this entails, until now the biological mechanisms behind this condition were unknown, due to the structural and functional complexity of the kidney.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p>Now, <span data-contrast=\"auto\">a study<\/span><span data-contrast=\"auto\"> co-led by Dr <a href=\"http:\/\/www.ibecbarcelona.eu\/member\/934\/N%C3%BAria+Montserrat+Pulido\/\" target=\"_blank\" rel=\"noopener noreferrer\">N\u00faria Montserrat<\/a>, ICREA Research Professor and Principal Investigator of the group Pluripotency for organ regeneration at IBEC, jointly with Dr <a href=\"https:\/\/www.med.upenn.edu\/susztaklab\/pi.html\" target=\"_blank\" rel=\"noopener noreferrer\">Katalin\u00a0Susztak<\/a> from the University of Pennsylvania, and Dr <a href=\"https:\/\/life.gist.ac.kr\/prog\/gsPerson\/genomics\/ALL\/view.do?siteCode=genomics&amp;mno=32786&amp;tmplId=template_b3&amp;personId=B000000032781Wk1fD7a\" target=\"_blank\" rel=\"noopener noreferrer\">Ji-Hwan Park<\/a> from the Korea Gwangju Institute of Science and Technology (GIST), reveals the genes responsible for chronic kidney disease and have shown that it is possible to reverse this deficiency, thus opening a possible therapeutic route against kidney damage. The identification was carried out using mini-kidneys created from human stem cells and generated in the laboratory using bioengineering techniques. In the study, published in the journal Cell Metabolism, they also demonstrated in animal models that it is possible to reverse this deficiency.\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Using innovative techniques such as single cell RNA-sequencing, the researchers\u00a0unraveled\u00a0the crucial role tha<\/span><span data-contrast=\"auto\">t lipid metabolism genes play in protecting against chronic kidney failure. For this, the\u00a0analyzes\u00a0were carried out with animal models and human\u00a0mini-kidneys. The results were validated using almost one hundred patient samples.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">In healthy adults, each of o<\/span><span data-contrast=\"auto\">ur kidneys contains an average of 1.5 million nephrons, the basic functional units of the kidney that constantly filter waste products from the blood. Among the different cells that make up these filtration units, the epithelial cells of the proximal tubul<\/span><span data-contrast=\"auto\">e of the nephron are responsible for reabsorbing water and solutes, representing 90% of the total renal mass. In almost all cases of chronic kidney disease, damage occurs in these cells, but until now, it was not known which cellular mechanisms were respon<\/span><span data-contrast=\"auto\">sible for such dysfunction.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">By sequencing the RNA of each of the thousands of cells present in the kidney, the researchers observed for the\u00a0<\/span><span data-contrast=\"auto\">first<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">time<\/span><span data-contrast=\"auto\">\u00a0important differences between healthy and diseased cells in the proximal tubule. Specifically, in the kid<\/span><span data-contrast=\"auto\">neys of mice with chronic kidney disease, they detected that a greater proportion of the proximal tubule cells had a different molecular signature than that found in the kidneys of healthy animals.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Noting these observations, the IBEC researchers went one s<\/span><span data-contrast=\"auto\">tep further and, using human\u00a0mini-kidneys, demonstrated that these changes were due to a\u00a0<\/span><span data-contrast=\"auto\">decrease in the expression of some of the genes that regulate lipid metabolism in proximal tubule cells.
